Nonalcoholic fatty liver disease (NAFLD) is a condition in which too much fat is stored in the liver. Nonalcoholic steatohepatitis (NASH) is a type of NAFLD marked by liver inflammation, which may progress to scarring and irreversible damage similar to that caused by heavy alcohol use. Components of glucose matabolism, insulin signaling, inflammation, oxidative stress, ER sgtress and apoptosis are all involved and interconnected in the progression of this disease.c6bb5cf1bb25b7fe96d4dc8ce67b06aacSER307hepatocyteCL:0000182Cell Ontologynonalcoholic fatty liver diseaseDOID:0080208Human Disease Ontologydisease pathwayPW:0000013Pathway Ontologyliver disease pathwayPW:0001413Pathway Ontology19352614PubMedMolecular mechanisms involved in NAFLD progression.J Mol Med (Berl)2009Malaguarnera MDi Rosa MNicoletti FMalaguarnera L22705006PubMedA liver full of JNK: signaling in regulation of cell function and disease pathogenesis, and clinical approaches.Gastroenterology2012Seki EBrenner DAKarin M16364488PubMedInsulin resistance, adiponectin, cytokines in NASH: Which is the best target to treat?J Hepatol2006Larter CZFarrell GC22570745PubMedNASH is an Inflammatory Disorder: Pathogenic, Prognostic and Therapeutic Implications.Gut Liver2012Farrell GCvan Rooyen DGan LChitturi S20303879PubMedEndoplasmic reticulum stress and the inflammatory basis of metabolic disease.Cell2010Hotamisligil GS12891546PubMedHepatocyte apoptosis and fas expression are prominent features of human nonalcoholic steatohepatitis.Gastroenterology2003Feldstein AECanbay AAngulo PTaniai MBurgart LJLindor KDGores GJ21145844PubMedEndoplasmic reticulum stress in liver disease.J Hepatol2011Malhi HKaufman RJ18242193PubMedMitochondrial dysfunction and oxidative stress in the pathogenesis of alcohol- and obesity-induced fatty liver diseases.Free Radic Biol Med2008Mantena SKKing ALAndringa KKEccleston HBBailey SM28983452PubMedRole of FXR in Liver Inflammation during Nonalcoholic Steatohepatitis.Curr Pharmacol Rep2017Armstrong LEGuo GL25501551PubMedMicro-RNA 21 inhibition of SMAD7 enhances fibrogenesis via leptin-mediated NADPH oxidase in experimental and human nonalcoholic steatohepatitis.Am J Physiol Gastrointest Liver Physiol2015Dattaroy DPourhoseini SDas SAlhasson FSeth RKNagarkatti MMichelotti GADiehl AMChatterjee S30315846PubMedSelonsertib (GS-4997), an ASK1 inhibitor, antagonizes multidrug resistance in ABCB1- and ABCG2-overexpressing cancer cells.Cancer Lett2018Loomba RLawitz EMantry PSJayakumar SCaldwell SHArnold HDiehl AMDjedjos CSHan LMyers RPSubramanian GMMcHutchison JGGoodman ZDAfdhal NHCharlton MR
